County Route 41 is a short north–south route located in the Town of Byron named South Holley Road. CR 41 starts its southern end at CR 14 (N. Bergen Rd.) and heads due north for just over a half-a-mile. There it reaches the county line where the Orleans CR 73 designation takes over. The roadway keeps its name north into Orleans County.

County Route 42 is an overall north–south route, however its starts out running east–west as Walkers Corners Road at the intersection of Genesee County Routes 19 & 19A. From there CR 42 heads west until it reaches Starowitz Road onto which it turns north. At 1.43 miles, CR 42 reaches CR 13 (Bank Street Rd.) where it is briefly broken, but resumes northeast of there 0.15 miles as Transit Road. As Transit Road, CR 42 heads due north for the remainder of its route; intersecting New York State Route 262 (Byron-Elba Rd.) at two and three-quarters miles, and ending at approximately 5.22 miles at Watson Road, where the country route designation ends.

CR 43, named Salt Road, is only a quarter of a mile long and runs from New York State Route 77/CR 12 (Lewiston Rd.) north a short distance to the Orleans County Line where Salt Road continues as Orleans CR 52.

County Route 45 is an east–west route in the Towns of Alexander and Darien named Attica Road. CR 46 begins its southern (or southeastern) end at New York State Route 238 just north of the Village of Attica. From here, CR 46 heads northwest crossing into the Town of Darien at almost nine-tenths of a mile at CR 36 (Alexander-Darien Townline Rd.). CR 46 continues northwest paralleling roughly to the south of NY 238, until it turns more directly north and ends at NY 238 at just over 2 miles.

County Route 46 is an east–west county route located in the Town of Batavia, north of the City of Batavia. CR 46 begins at itys west end at New York State Route 98 (Oak Orchard Rd.) named East Saile Drive. At 0.71 miles, CR 46 crosses State Street Road and changes its name to West Saile Drive. As the roadway heads east, it passes along the southern edge of the Genesee County Airport for much of the rest of its length, before ending at CR 13 (Bank Street Rd.)

CR 48 starts its west end at CR 13 (Bank Street Rd.) and heads west , passing south of Genesee Community College before reaching its end at CR 19 (Batavia-Stafford Townline Rd.) on the Batavia-Stafford Town Line.

County Route 49 is a fairly short east–west route named Raymond Road in the Town of Bethany. CR 49 starts its west end at the intersection of Linden and Bethany Center Roads and at CR 15. (CR 15 runs west along Linden and north along Bethany Center). CR 49 heads east from here running along the northern border of the Genesee County Park and Forest [1]. Memory Lane (CR 52) is the only road CR 49 intersects and allows access to the park. At 1.49 miles, CR 49 recahes it eastern end at CR 35 (East Rd.).

County Route 50 is a north–south county route located in the Towns of Pavilion and Le Roy named Perry Road. Route 50 starts it southern end just north of the Wyoming County Line at an intersection with New York State Route 63 and the northern terminus of New York State Route 246 (Perry Rd.). From here, CR 50 heads due north through Pavilion, running parallel to the east of New York State Route 19 for its whole length. At just over 3 miles, CR 50 intersects US 20 (Broadway), and continues heading north, into the Town of Le Roy. At almost 6.4 miles CR 50 comes to its north end at NY 19 (Warsaw Rd.), southwest of the Village of Le Roy.

County Route 51 is a north–south county route primarily located within the Town of Darien and crosses into the Town of Pembroke before ending. CR 51 is named Colby Road for its entire length and begins at a four-way intersection with US Route 20 (Broadway) and Tinkham Road. CR 51 heads due north from here intersecting only local and county roadways, and crossing into the Town of Pembroke just before it reaches New York State Route 33 (Genesee St.) where the country route has its north end.

County Route 52 is the highest numbered route in the county. It is an east–west route located within the Town of Bethany named Memory Lane. CR 52 is a county route that runs through the Genesee County Park and Forest [2]. It starts its west end at Bethany Center Road, just south of the Bethany Center Road and Raymond Road (CR 49) intersection. CR 52 heads east into the park, and turns south to run through the southern section and then turns back to the north to its exit at CR 49 (Raymond Rd.). In all, CR 52 is just under 2 miles in length.
